Conformational Transitions Visualized in Single Polyelectrolyte Molecule Afm Experiments

چکیده
Introduction High resolution AFM allows for the visualization of the adsorbed polymer chains. If the chains are rapidly trapped by the substrate the twodimensional structures visualized with AFM reflect the sample prehistory in solution. In contrast to scattering methods the single molecule AFM experiments allow for the investigation of the molecules deposited from very diluted solutions when the interchain interactions can be neglected. It is of special interest for polyelectrolyte systems if the interchain interactions may cause various technical problems in scattering experiments.

Molecular processes studied at a single-molecule level using DNA origami nanostructures and atomic force microscopy.
DNA origami nanostructures allow for the arrangement of different functionalities such as proteins, specific DNA structures, nanoparticles, and various chemical modifications with unprecedented precision. The arranged functional entities can be visualized by atomic force microscopy (AFM) which enables the study of molecular processes at a single-molecular level. Examples comprise the investigat...
متن کاملMean field theory for the intermolecular and intramolecular conformational transitions of a single flexible polyelectrolyte chain.
The diMarzio theory has been extended to elucidate the intermolecular and intramolecular phase segregations of a single flexible chain polyelectrolyte in dilute salt-free solutions. At the long chain limit, this theory yields the formalism obtained from the more sophisticated Edward Hamiltonian for polyelectrolyte problems. The calculated phase diagram exhibits the features of a first-order pha...
متن کاملChair-boat transitions in single polysaccharide molecules observed with force-ramp AFM.
Under a stretching force, the sugar ring of polysaccharide molecules switches from the chair to the boat-like or inverted chair conformation. This conformational change can be observed by stretching single polysaccharide molecules with an atomic force microscope. In those early experiments, the molecules were stretched at a constant rate while the resulting force changed over wide ranges. Howev...
متن کاملInvestigating single molecule adhesion by atomic force spectroscopy.
Atomic force spectroscopy is an ideal tool to study molecules at surfaces and interfaces. An experimental protocol to couple a large variety of single molecules covalently onto an AFM tip is presented. At the same time the AFM tip is passivated to prevent unspecific interactions between the tip and the substrate, which is a prerequisite to study single molecules attached to the AFM tip. Analyse...
متن کاملComputer modeling of charged polymers
We give a short introduction into polyelectrolytes and show some of the problems of modeling charged polymer systems. We concentrate on two model systems. The first one consists of flexible polyelectrolytes exposed to a poor solvent. These polymers show interesting conformational behavior. We will discuss them and hint to the difficulties of observing conformational transitions in light scatter...
